What is MOTS-c?

MOTS-c is a 16-amino-acid peptide encoded in the mitochondrial 12S rRNA gene. It was described in 2015 by Pinchas Cohen’s group at USC.

It acts on metabolism, in part through AMPK.

Lee and colleagues reported in Cell Metabolism (2015) that MOTS-c prevented diet-induced obesity and insulin resistance in mice.

Reynolds and colleagues showed in 2021 that it improved physical capacity in young, middle-aged and old mice.

How to mix a MOTS-c vial

The vial holds MOTS-c as a dry, freeze-dried cake. To use it, dissolve it in bacteriostatic water, letting it run slowly down the side of the glass so it doesn’t hit the powder directly. Swirl or roll the vial until it’s clear. Shaking can damage peptides, so skip it.

The amount of water you add sets the concentration. For example, adding 2mL of bacteriostatic water to a 10mg vial gives 5mg per mL. Write the volume and the date on the label so anyone picking up the vial later knows what’s in it.

We sell bacteriostatic water in 30mL vials, and acetic acid solution for the few peptides that don’t dissolve well in water.

Choosing a MOTS-c vial size

The vial comes in 10mg and 40mg. The contents are the same apart from the amount, so the choice comes down to how much your study needs and how quickly you’ll use a mixed vial.

Mixed peptides don’t last as long as dry ones, so several smaller vials can work out better than one large one if you’re running a slow study.

How we test MOTS-c

We test each batch of MOTS-c by LC-MS, which pairs liquid chromatography with mass spectrometry. The chromatography splits the sample into its components and the mass spectrometer identifies them, which tells you both identity and purity. This batch came in at 99% or higher.

Small differences between batches are normal, so write the batch number from the label into your records. If something looks wrong when your order arrives, like discoloured powder or a cloudy solution, set it aside and don’t use it.

How to store MOTS-c

Keep it in the fridge at 2 to 8°C and don’t freeze it. Once mixed, keep the vial in the fridge and use it within a few weeks. Unmixed vials keep much longer, so only reconstitute what you need. Label anything you decant with the compound, amount and date, and keep it out of reach of children and pets.
