What is Idebenone?

Idebenone is a synthetic analog of coenzyme Q10 with a shorter tail, first developed by Takeda. It can carry electrons past complex I in the mitochondrial chain.

It’s approved in Europe as Raxone for Leber’s hereditary optic neuropathy.

Klopstock and colleagues reported in Brain (2011) on the RHODOS trial, which showed a trend toward better vision in LHON.

Earlier trials in Alzheimer’s disease didn’t show a clear effect.

To make a stock solution, decide on the final volume and concentration first. For 30mL at 10mg/mL you’d weigh 300mg of Idebenone, dissolve it in about 25mL of solvent, then top up to exactly 30mL.

Check the compound’s solubility in your chosen solvent before you start, and try a small test amount first. Store the finished solution in a labelled glass bottle.

Every batch of Idebenone is checked with liquid chromatography-mass spectrometry (LC-MS) before it goes on sale. LC-MS separates a sample into its parts and identifies each one by mass, so it confirms what the compound is and how much of the sample it makes up. The current batch tested at 99% or higher.

Small differences between batches are normal, so write the batch number from the label into your records. If something looks wrong when your order arrives, like discoloured powder or a cloudy solution, set it aside and don’t use it.
